Proteolytic bioactive form of Wnt5a is related to the pathogenesis of IBD in view of mucosal healing and anti-inflammation | |||

Background and aims: We have reported about the role of Wnt5a peptide secreted from colonic myofibroblast to enhance colonic epithelial proliferation (Sci Rep. 2016). We have also found that Wnt5a mRNA is highly induced in colonic mucosa in the patients of severe ulcerative colitis. Therefore, we speculated that Wnt5a may have an important role in the pathogenesis of ulcerative colitis. In the present study, we investigated the role of Wnt5a peptide to enhance colonic epithelial restitution and anti-inflammatory effect, which those are very important to control condition of the patients with IBD. Material and methods: Cultured Young Adult Mouse Colonic (YAMC) epithelial cell is used to investigate the enhancement of restitution by recombinant Wnt5a peptide (36aa). In vivo studies to investigate the role of Wnt5a peptide to enhance colonic epithelium were conducted using recovery phase of mouse DSS colitis model by intra peritoneal injection of Wnt5a peptide. In the respective experiment, Wnt5a peptide was administered at the same time of DSS treatment to investigate the effect about the reduction of colonic inflammation. Results: The restitution of YAMC cells was enhanced by the treatment of Wnt5a peptide. The recovery of DSS colitis model was enhanced by the treatment of Wnt5a peptide. The colonic inflammation induced by DSS was also reduced by Wnt5a peptide via the reduction of inflammatory cytokines such as TNF-α, KC, and IFN-γ. Conclusion: Wnt5a peptide is thought to play an important role of colonic epithelial restitution and anti-inflammatory effect of colitis. These results implicate the possibility for clinical application to regulate intestinal epithelial wound healing and colonic inflammation. |
