Anti-αvβ6 integrin antibody for diagnosis of ulcerative colitis: A nation-wide multicenter study

Ulcerative colitis (UC) is an idiopathic, chronic, and inflammatory disorder that mainly affects the colorectum, and has no established diagnostic biomarker. We recently reported anti-αvβ6 integrin antibody with high sensitivity and specificity for diagnosing UC which were further confirmed in a Japanese pediatric population, a Swedish cohort, and US cohorts. However, there is no large-scale validation study on this antibody. Therefore, we conducted a nation-wide, multicenter study on anti-αvβ6 integrin antibody for diagnosis of UC (UCV6-Dx study).
In this study, 28 Japanese high-volume referral centers participated. Patients aged 18 years or older with UC, Crohn's disease, and other gastrointestinal diseases including colon cancer, infectious enterocolitis, drug-induced colitis, and intestinal Behcet's disease were enrolled. The target numbers of participants with UC, Crohn's disease, and other gastrointestinal diseases were more than 1000, 500, and 300, respectively. The primary outcome was the diagnostic value of anti-αvβ6 integrin antibody. Secondary outcomes were factors associated with false negative results in UC patients and false positive results in non-UC patients, and diagnostic value of each facility.
